Oil Immersion Objective: A Supervised Workflow

Confirm that the objective is designed for oil
Use an oil-immersion objective only under instructor supervision and the microscope manufacturer's procedure. Read the objective label and laboratory instructions. Never place oil on a dry objective, an ordinary coverslip setup, or a slide not approved for the method. An observation made through this procedure must never be used to diagnose a person, animal, infection, or disease; clinical interpretation belongs to a qualified pathologist through the institution's procedure.
Use only the specified immersion medium. Read the Safety Data Sheet (SDS) for the specific product and follow its protective-equipment, exposure-response, and disposal requirements together with laboratory procedure. Oil can damage incompatible objectives and contaminate the nosepiece, stage, slides, and nearby lenses.
Focus and center before adding oil
Find the approved prepared specimen at low power through the standard focusing workflow. Move to the lab's designated high dry objective, focus with fine control, and center the exact target. A target near the edge will probably vanish under oil because the field becomes smaller.
Do not let a dry objective touch oil left on the slide. Follow the instrument's objective-rotation sequence exactly.
Apply the approved amount
Place the specified small amount of immersion oil on the approved coverslip area without touching the applicator tip to glass or specimen. Keep the bottle capped when not in use and prevent spills.
Rotate the oil objective into position through the instructed path so its front lens contacts the oil without crossing another objective through the drop. Watch from the side as required. Stop if clearance, labeling, or alignment is uncertain.
Focus only through the approved control
Look through the eyepiece and use fine focus as instructed. Do not use coarse focus with the oil-immersion objective. Never sweep the objective toward the slide; use only the approved fine-focus procedure. Adjust illumination for the oil objective through the lab setup; high brightness alone cannot rescue an uncentered or poorly prepared target.

Prevent cross-contamination
Do not rotate a dry objective into the oil. If that occurs, stop and report it rather than wiping every lens with the nearest tissue. Use only approved lens materials and cleaning fluid, and keep oil away from fingers, eyes, clothing, and electrical parts.
Follow the laboratory's spill and exposure procedure. Do not use household solvents or unapproved alcohols; they may damage coatings, adhesives, plastics, or finishes.
Clean immediately after use
At the end, rotate away through the approved route and remove oil from the immersion objective and slide according to the manufacturer and lab method. The microscope cleaning guide covers lens paper, order, inspection, and storage.
Do not leave oil to dry on the objective. Dispose of contaminated materials through the specified route, wash hands as required, and document any damage or spill.
Before leaving the bench, confirm the oil bottle, contaminated materials, objective, slide, stage, and work surface have all reached their required clean or stored state.
Record the full setup
Write down the instrument, eyepiece, oil objective, specimen, medium, illumination setup, and any image issue. Oil immersion is not simply “more zoom.” It is a matched optical procedure whose value depends on the correct objective, medium, slide, focus, cleaning, and supervision.
